چکیده

Transport of the organic molecule malachite green, across a bilayer of dioleoylphosphatidylglycerol liposome in aqueous Ž . solution, has been observed using a new method, namely second harmonic generation SHG . Following the rapid mixing of liposome and malachite green solutions, a decay of the SHG signal in time is observed as the malachite green bound to the outer liposome surface crosses to the inner liposome surface. The effect of liquid-crystalline versus gel liposome structures on the transport across the bilayer is reported also. q 1998 Elsevier Science B.V. All rights reserved.

Effects of bilayer surface charge density on molecular adsorption and transport across liposome bilayers.

Second harmonic generation (SHG) was used to study both the adsorption of malachite green (MG), a positively charged organic dye, onto liposomes of different lipid compositions, and the transport kinetics of MG across the liposome bilayer in real time. We found that the dye adsorption increased linearly with the fraction of negatively charged lipids in the bilayer. Similarly, the transport rate...

Effect of cholesterol on molecular transport of organic cations across liposome bilayers probed by second harmonic generation.

The effect of cholesterol on the molecular transport of an organic cation, malachite green (MG), across large unilamellar dioleolyphosphatidylglycerol (DOPG) liposome bilayers with 0-50 mol% cholesterol was studied by second harmonic generation (SHG). Because SHG is a surface-specific technique, it requires no labeled molecule, quencher, or shifting agent to distinguish the location of the solu...

Antibiotic assisted molecular ion transport across a membrane in real time.

The transport of an organic cation across a 4-5 nm liposome bilayer is observed in real time using second harmonic generation. It is proposed that an electrostatic barrier between the inside and outside of the liposome develops as the cation crosses the bilayer. This would explain why the SHG signal does not approach zero at long times. To test this mechanism, the antibiotic valinomycin, which ...

Second harmonic studies of ions crossing liposome membranes in real time.

The transport kinetics of the positively charged triphenylmethane dye, malachite green (MG(+)), across liposome bilayers effects the transport of monovalent inorganic cations when ionophores are present in the membrane. Three different types of ionophores characterized by different transport mechanisms have been studied. The ionophores are gramicidin A (gA) (a channel former), valinomycin (VAL)...

Catalytic photoinduced electron transport across a lipid bilayer mediated by a membrane-soluble electron relay.

Unidirectional photocatalytic electron transfer from a hydrophilic electron donor encapsulated in the interior of a liposome, to a hydrophilic electron acceptor on the other side of the membrane, has been achieved using the simple membrane-soluble electron relay 1-methoxy-N-methylphenazinium (MMP(+)). The total amount of photoproduct (>140 nmol) exceeds the number of moles of MMP(+) present (12...
